CONSOLIDATE DNA FINGERPRINTING REPORT

Project title: DNA Fingerprinting 38 crops with 5 to 8 RAPD markers each.

Introduction:

In collaboration with Mr. Mahendra Kumar Trivedi, Bangalore Genei carried out a DNA mapping project. The aim of this project was to observe the impact of Mr. Mahendra Kumar Trivedi's treatment modality on various groups of crop lines and to analyze the percentage of DNA Polymorphism (Mutation) observed between Treated and Untreated samples of 38 crops.

The selection of the crop lines & the treatment part of the said project was designed and carried out by Mr. Mahendra Kumar Trivedi (Mumbai) himself. Our laboratory was used to detect the polymorphism (mutation) on consultancy basis.

Dr Avijit Roy (Manager - R&D) & Mr. Mehul Rajgor (Designation - Assistant Executive Manager - Sales) conducted this project.

Objective:
The objective of this project was to genotype the 2 samples (Treated vs. Untreated) of the same line with 5 to 8 RAPD markers as per the requirement of individual crop.

Duration of the project: 3 months

Setting:

Samples:
Both Control (untreated) and Treated samples were procured from Mr. Mahendra Kumar Trivedi in two separate groups.

Group A - Untreated (Control) Samples (Series 1) - 38 samples
Group B - Treated Samples (Series 2) - 38 samples

All varieties of Control and Treated seeds were sowed in the soil separately. After germination when plants reached the appropriate stage, leaf discs were harvested from each plant. DNA extractions were performed according to standard CTAB method (Sambrook et al; 2000). Bangalore Genei selected Markers of each crop as per the requirement of individual crop and PCR reactions were amplified following Bangalore Genei standard protocol for RAPD on all samples.

The percentage of the true Polymorphism (Mutation) observed in Control and Treated samples have been recorded in a tabular form as follows:

Conclusion:
The treatment modality of Mr. Mahendra Kumar Trivedi is not known to us, the company is only concerned about detecting the DNA Polymorphism (Mutation) in Control & Treated lines of 38 crops.
