PROCEDURES FOLLOWED FOR
 
IAA EXTRACTION & ANALYSIS
 
  • IAA was analyzed using Tang and Bonner's method
  • Freshly prepared Salkowski's reagent was used for detection
  • 200 mg plant tissue was grinded with 5 ml of 80% chilled methanol
  • Extract was filtered through Whatmann paper no. 1
  • Final volume of extract was made up to 10 ml using 80% chilled methanol
  • Optical density was measure after 30 minutes at 530 nm.
 
GLUTATHIONE EXTRACTION
 
The glutathione extraction procedure was as follows:
  • 5 gm of leaves was crushed in 5 ml of 80% chilled methanol (as a solvent)
  • Extract was sonicated for 10 min.
  • After sonication the total volume was adjusted to 5 ml.
  • Then 1ml of 5% tricholoroacetic acid was added to the extract
  • This sample was used for glutathione analysis.
GLUTATHIONE ANALYSIS
 
  • The glutathione levels were estimated by Moron et.al's (1979) method.
  • Trichloroacetic acid was taken as blank.
  • Procedure given in Biochimica et Biophysica acta, 582, 1979, 67-78 was followed.
INFERENCE
Very significant increase in germination, seedling growth associated with increased level of auxin (IAA) in both root and shoot of seedlings of blessed plants is 13% &16.60% respectively which suggests an early establishment of healthy plant.
 
Absence of any type of disease (i.e. viral, fungal and insect) was noticed in blessed plants, which is supported by 206.71% increased level of glutathione in them.
